سنجش میزان بارگیری و رها سازی دو پوشش انتریک برای انتقال اسید نوکلئیک از طریق دهان

<p style="text-align: justify"><strong>زمینه و هدف</strong>: ژن درمانی شیوه نوین و نوید بخش درمانی است که انتقال کارآمد ژن مهمترین مرحله آن محسوب میگردد. علاوه بر این، انتقال به گونهای غیرتهاجمی نیز در مرحله به کارگیری این روش اهمیت فراوان مییابد. تجویز دهانی روشی است که بسیار مورد اقبال بیماران قرار گرفته است. برای انتقال دهانی دارو، روشهای متعددی به کار گرفته شدهاند که در همه آنها میزان بارگیری و رهاسازی در هدفمند ساختن و کارایی انتقال بسیار موثر است. در این تحقیق بسته بندی ذرات ژنی در دو نوع پوشش انتریک و همچنین مقایسه میزان بارگیری و رهاسازی ماده محصور شده این دو پوشش، در شرایط آزمایشگاهی صورت پذیرفت.</p>
<p style="text-align: justify"><strong>مواد و روشها</strong>: در این مطالعه تجربی ابتدا DNA به روش coacervation با کیتوزان ترکیب و سپس پلی پلکسهای حاصل شده با روش تبخیر حلال، پوشش دهی شدند. به منظور بررسی تشکیل و رفتار ذرات از روش FTIR و دو محلول با pHهای دو حاشیهی PKa پوشش انتریک استفاده شد.</p>
<p style="text-align: justify"><strong>یافتهها</strong>: ذرات تشکیل شده، پایداری مشابه درpH پایین دارند و تفاوت قابل توجهی نشان نمیدهند. میزان رهاسازی یودراجیت L100 نسبت به L100-55 آهستهتر و تدریجی است. یودارجیت L100 قابلیت بهتری در میزان بارگیری DNA نشان میدهد.</p>
<p style="text-align: justify"><strong>نتیجهگیری</strong>: بر اساس رفتار دو نوع پارتیکل تشکیل شده، در تجویز دهانی ژن میتوان از یودراجیت L100-55 با هدف ترنسفکشن ابتدای روده کوچک و از یودراجیت L100 برای ترنسفکشن سطح وسیعتری از ابتدا تا انتهای روده کوچک و کولون استفاده کرد.</p>
<p dir="ltr" style="text-align: justify width: 733px height: 228px"><span style="font-size: 14px"><span style="font-family: times new roman"><strong><em>Background</em></strong>: Gene therapy is a recent promising treatment that effective gene transfer is considered as its most important step. Furthermore noninvasive method of transfer will be important to, when gene therapy is supposed to be applied. Administration of drugs in oral rout is more appreciated by patients. Loading and release rates are very important in targeting and effectiveness of transfer in all different methods that have been used for oral drug transfer. Here, we have studied packing of gene particles into two different enteric coats and compared these two coats in loading of entrapped materials and there release rate in vitro.</span></span></p>
<p dir="ltr" style="text-align: justify width: 733px height: 228px"><span style="font-size: 14px"><span style="font-family: times new roman"><strong><em>Materials and Methods</em></strong>: First, DNA was mixed with chitosan by coacervation technique and resulted polyplexes were coated using solvent evaporation technique. FTIR and two different pHs, less and more but near eudragit pKa, were used to evaluate formation of particles and their behavior.</span></span></p>
<p dir="ltr" style="text-align: justify width: 733px height: 228px"><span style="font-size: 14px"><span style="font-family: times new roman"><strong><em>Results</em></strong>: Formed particles have similar stability in low pH and their differences are trivial. Eudragit L100 release rate is really slower than L100-55 and gradual. Eudragit L100 shows better ability in loading rate. </span></span></p>
<p dir="ltr" style="text-align: justify width: 733px height: 228px"><span style="font-size: 14px"><span style="font-family: times new roman"><strong><em>Conclusion</em></strong>: According to two formed particles' behavior, eudragit L100-55 might be used in oral gene transfer targeting of initial part of small intestine and eudragit L100 might be used for wider surface of small intestine, from the initial to the end part, and colon.</span></span></p>
