بررسی پاسخ تکثیر لنفوسیتی در مبتلایان به بروسلوز حاد و مزمن

<p dir="rtl" style="text-align: justify; margin-right: 28.9pt"><strong>زمینه و هدف:</strong> بروسلوز یک باکتری داخل سلولی است که سبب عفونت مزمن در انسان و حیوانات اهلی میشود. مکانیسمهای مزمن شدن این بیماری پیچیده و ناشناخته هستند. پاسخ های ایمنی ممکن است در مزمن شدن این بیماری نقش داشته باشند. در این مطالعه پاسخ تکثیر لنفوسیتی در بیماران مبتلا به بروسلوز حاد و مزمن مورد ارزیابی قرار گرفت.</p>
<p dir="rtl" style="text-align: justify; margin-right: 28.9pt"><strong>مواد و روش</strong><strong>ها:</strong> این مطالعه توصیفی-تحلیلی، بر روی 22 بیمار مبتلا به بروسلوز حاد، 21 بیمار مبتلا به بروسلوز مزمن و 21 فرد سالم که از نظر سن، جنس و زمینه ژنتیکی با بیماران انطباق داشتند، انجام گرفت. لنفوسیتهای خون محیطی با محیط گرادیان فایکول جدا شدند و میزان تکثیر سلولی در حضور آنتی ژن و فیتوهماگلوتنین-آ در شرایط کشت با روش <span dir="ltr">MTT</span> اندازهگیری شد.</p>
<p dir="rtl" style="text-align: justify; margin-right: 28.9pt"><strong>یافته</strong><strong>ها: </strong>اندیکس تحریک اختصاصی آنتی ژنهای بروسلا در بیماران مبتلا به بروسلوز مزمن به طور معنیداری در مقایسه با بیماران مبتلا به فرم حاد بیماری کمتر بود(001/0=<span dir="ltr">p</span>). همچنین در تحریک لنفوسیتی با فیتوهماگلوتنین-آ نشان داده شد که پاسخ تکثیری در بیماران مزمن نسبت به گروه کنترل کمتر میباشد(04/0=<span dir="ltr">p</span>).</p>
<p style="text-align: justify; margin-right: 40px"><strong><span dir="rtl">نتیجه</span></strong><span dir="rtl"></span><strong><span dir="rtl">گیری</span></strong><span dir="rtl">: نتایج نشان داد که کاهش پاسخ تکثیر لنفوسیتی در بیماران مبتلا به برسلوز مزمن ممکن است به دلیل القاء پدیده آنرژی در این بیماران باشد.</span></p>
<p style="text-align: justify"><strong><em>Background:</em></strong> Brucella is an intracellular bacterium that causes chronic infection in humans and domestic animals. The underlying mechanisms that cause prolonged illness are complex and not fully understood. Immune responses may have an important role in the chronicity of infection. Here, we evaluated the lymphocyte proliferation responses in patients with chronic and acute brucellosis.</p>
<p style="text-align: justify"><strong><em>Materials and Methods:</em></strong> This descriptive - analytical study was performed on 22 patients with acute brucellosis, 21 patients with chronic brucellosis and 21 healthy people with the similar age, sex and genetic background as control group. Peripheral lymphocytes were isolated using Ficoll and the cellular proliferation was quantified in presence of antigen and phytohemaglutinin-A by MTT method.</p>
<p style="text-align: justify"><strong><em>Results:</em></strong> The brucella antigen-specific stimulation index in patients with chronic brucellosis was significantly lower than the acute brucellosis patients (p=0.001). Also, stimulating the lymphocytes with phytohemaglutinin-A has shown that proliferative response in patients with chronic brucellosis was lower than the other groups (p=0.04).</p>
<p style="text-align: justify"><strong><em>Conclusion:</em></strong> The results indicated that chronic brucellosis inhibits lymphocyte proliferation. This inhibition of lymphocyte proliferation may be due to the induction of anergy.</p>
