fa اثربخشی عصاره‌های ترکیبی آویشن معمولی و شنبلیله در مدل ریسه‎ کاندیدا آلبیکانس و کاهش بیان ژن آسپارتیل پروتئیناز ترشحی 1 Efficacy of Thymus vulgaris and Trigonella foenum-graecum Extracts Alone and in Combination in Candida albicans Hyphae Model and in Downregulation of Secreted Aspartyl Proteinase 1 Gene Expression علوم پایه Basic Sciences پژوهشي اصیل Original Atricle مقدمه: ظهور مقاومت ضدقارچی در کاندیدا آلبیکانس یک تهدید جدی برای سلامت جهانی است. این امر، توسعه رویکردهای درمانی جدید را ضروری می‌سازد. مطالعه حاضر به بررسی اثرات ضدقارچی و ضد ریسه&lrm;ای عصاره‌های آویشن معمولی (Thymus vulgaris) و شنبلیله (Trigonella foenum-graecum)، به&lrm;تنهایی و ترکیبی، بر کاندیدا آلبیکانس می‌پردازد. روش کار: عصاره گیاهان آویشن معمولی و شنبلیله با آب داغ (60 درجه سلسیوس) و استخراج سوکسله با متانول (10 درصد) تهیه شد. آزمون حساسیت کاندیدا آلبیکانس با استفاده از روش‌های دیسک دیفیوژن و میکرودایلوشن براث طبق دستورالعمل‌های مؤسسه استاندارد آزمایشگاهی و بالینی (CLSI) انجام شد. مدل ریسه در حضور عصاره&lrm;های الکلی آویشن معمولی و شنبلیله به‌تنهایی و در ترکیب ایجاد شد. مهار رشد ریسه از طریق رنگ‌آمیزی کریستال ویولت، مشاهده میکروسکوپی و تجزیه و تحلیل بیان ژن SAP1 ارزیابی شد. یافته‌ها: ۹۰ درصد از جدایه‌های کاندیدا آلبیکانس مورد آزمایش، مقاومت به فلوکونازول را نشان دادند. ترکیب عصاره‌های آبی و الکلی آویشن با عصاره‌های شنبلیله اثرات هم&lrm;افزایی، هم&lrm;افزایی جزئی و افزایشی ضدقارچی را نشان داد. به&lrm;طور خاص، عصاره‌های الکلی آویشن و شنبلیله، به&lrm;تنهایی و در ترکیب، فعالیت ضد ریسه&lrm;ای قابل&lrm;توجهی را نشان دادند. این فعالیت با کاهش درصد ریسه‌های تشکیل شده، کاهش تعداد سلول‌های پلانکتونی، مهار تبدیل سلول&lrm;های پلانکتونی به ریسه و کاهش بیان ژن SAP1 مشخص شد. نتیجه‌گیری: این یافته‌ها نشان می‌دهد که عصاره‌های آویشن، به ویژه در ترکیب با شنبلیله، به عنوان یک رویکرد جایگزین امیدوارکننده برای مبارزه با عفونت‌های مقاوم کاندیدا آلبیکانس و مهار رشد ریسه مطرح می‌شوند. فزون‌بر این، ژن SAP1 می‌تواند به عنوان یک هدف بالقوه برای اثر هم&lrm;افزایی مشاهده شده بین عصاره‌های الکلی آویشن و شنبلیله علیه ریسه‌های کاندیدا آلبیکانس در نظر گرفته شود. Introduction: The emergence of antifungal resistance in Candida albicans diseases poses a threat to global public health. New treatments are needed to target C. albicans and its ability to produce hyphae. The aim of this study was to investigate the potential synergistic effects and antifungal properties of Thymus vulgaris and Trigonella foenum-graecum extracts alone and in combination on C. albicans. Methods: In this experimental study, extracts of Thymus vulgaris and Trigonella foenum-graecum were prepared using hot water (60 °C) and Soxhlet extraction with methanol (10%). Yeast susceptibility testing was performed according to the Clinical and Laboratory Standards Institute disk diffusion and broth microdilution guidelines. The hyphal model was created in the presence of alcoholic extracts of Thymus vulgaris and Trigonella foenum-graecum alone and in combination. Crystal violet staining, microscopic observation and gene expression analysis were used to evaluate the inhibition of hyphal growth. Results: The results showed that 90% of the C. albicans isolates were resistant to fluconazole. Aqueous and alcoholic extracts of Thymus vulgaris in combination with Trigonella foenum-graecum showed synergistic, partially synergistic and additive effects. Alcoholic extracts of Thymus vulgaris with Trigonella foenum-graecum alone and in combination have anti-hyphae activity by reducing the percentage of hyphae, reducing the number of planktonic cells and the transition of planktonic cells to hyphae, and down-regulating the Secreted Aspartyl Proteinase 1 (SAP1) gene. Conclusions: Taken together, these results indicate that extracts of Thymus vulgaris alone and in combination with Trigonella foenum-graecum may offer a potent alternative strategy to combat resistant C. albicans infections and their ability to reduce hyphae formation.
