Journal of Arak University of Medical Sciences J Arak Uni Med Sci 1735-5338 2008-644X 2018 21 2 Gengenotypic Evaluation of Hsa-miR-433-3p Binding Site in the Regulatory Region of TYMS in Breast Cancer Patients Ali Arash Anoushirvani a.aa.arak2017@gmail.com Azam Ahmadi azam.ahmadi@modares.ac.ir Reza Aghabozorgi dr.aghabozorgi@arakmu.ac.ir Sara Khalili sarakhalili89@yahoo.com Maryam Sahraei sahraeimaryam@gmail.com Taha Fereydouni t.fereydouni@arakmu.ac.ir Zoha Khademi z.khademi@arakmu.ac.ir Abstract Background: Breast cancer is the most common cancer in women. It has been proven the association of cause of this disease with changes in several genes. One of the pathways associated with breast cancer is the folate reuptake pathway. The key enzyme of this pathway is coded by the TYMS gene. MicroRNAs control the expression of genes by binding to their regulatory regions. In this study, we evaluated changes in the regulatory region of TYMS gene with demographic characteristics (including the grade of cancer and metastasis) in breast cancer patients. Materials and Methods: In this study, the regulatory region of TYMS gene was investigated using related bioinformatics software. After collecting cancerous samples and DNA extraction from blood samples of normal and patients, change in the miRNA binding region by digestion with NlaIII enzyme was assayed. Results: Bioinformatics studies showed that the restriction site of some of the endonuclease enzymes in the 3'-UTR of the TYMS gene is related to the binding region of miRNAs, including Hsa-miR-433-3p. The results indicated the correctness of the genomic purification process, the PCR and enzymatic digestion reaction. In this study, in the regulatory region, CC homozygote, AC heterozygote and AA mutant homozygote variant had differences with control group (OR: 1.3465, %95 CI: 0.7275 to 2.4923, p<0.05). Also, the association of AA genotypes with metastasis and high grade of the patients was confirmed statistically. Conclusion: Studies have shown that some of polymorphisms in the key genes involved in cancer are directly related to their diagnosis and treatment process, and given the importance of timely diagnosis of cancer, the achievement of diagnostic biomarkers in breast cancer in the early stages will be important. Probably, the nucleotide change at the site of the microRNA binding site could be used as a diagnostic biomarker for degree of tumor progression. Journal of Arak University of Medical Sciences J Arak Uni Med Sci 1735-5338 2008-644X 2018 21 2 Investigation the Effect of Micelle Nanoparticles Containing Curcumin on Ciprofloxacin Resistant Isolates of Pseudomonas Aeruginosa and on mexC and mexD Genes Expression Bozorgmehr Imani Pirsaraei ramtin_imani@yahoo.com Najmeh Ranji n_ranji@iaurasht.ac.ir Leila Asadpour l.asadpour@yahoo.com Abstract Background: Pseudomonas aeruginosa is an opportunistic gram-negative bacterium that is a major cause of nosocomial infections such as severe burns. Curcumin is the main component of turmeric (Curcuma longa) that has anti-cancer and anti-inflammatory effects. The aim of this study was to evaluate antibacterial effect of curcumin in ciprofloxacin resistant isolates of Pseudomonas aeruginosa through mexC and mexD gene expression. Materials and Methods: In this descriptive-analytical study, Pseudomonas aeruginosa strains were obtained from hospitals and laboratories in Guilan province. After disc difusion and MIC tests, four ciprofloxacin resistant strains of Pseudomonas aeruginosa were treated by ciprofloxacin (1/2MIC) only (control sample) and in the combination with curcumin encapsulated in micelle nanoparticles (test sample). After 24h, RNA extraction and cDNA synthesis was performed. Then, the expression of mexC and mexD genes was evaluated quantitatively by Real-time PCR method in curcumin treated and un-treated cells Results: This study showed that combination of ciprofloxacin (1/2 MIC) with curcumin encapsulated in micelle nanoparticles led to approximately 50% of growth inhibition in Pseudomonas aeruginosa. In treated cells with curcumin and ciprofloxacin compared to treated cells with ciprofloxacin alone, mexC and mexD genes were significantly (p<0.05) downregulated >0.65 fold in three isolates and >0.1 fold in four isolates, respectively. Conclusion: Our results suggested that curcumin encapsulated in micelle nanoparticles combined with 1/2 MIC value of ciprofloxacin inhibits the growth of Pseudomonas aeruginosa through reducing mexC and mexD expression. Interval training, on the other hand, through reducing the adverse effects of diabetes, exerts a positive impact on diabetic individuals. The aim of the present study was to examine the effects of ten weeks of High Intensity Interval Training (HIIT) on reproductive hormones and sperm parameters in Wistar rats with diabetes mellitus type 2. Materials and Methods: In this experimental study, 36 Wistar rats with mean weight of 200±48 were randomly assigned to healthy control, diabetic control and diabetic + high intensity interval training groups. The diabetic training group received ten weeks of HIIT training by treadmill following the induction of diabetes. Twenty-four hours after the last training session, left epididymis of the rats was examined for studying sperm parameters and blood serum samples were examined for evaluating reproductive hormones. Data were analyzed by one-way ANOVA and Tukey's post hoc test at a significant level of 0.05%. Results: Ten weeks of HIIT training reduces fasting blood glucose (p=0.001) and significantly increases serum testosterone (p=0.001), LH (p=0.042) and FSH (p=0.024) levels in the HIIT training group in comparison to the diabetic group. In addition, sperm parameters (sperm count, survival rate and motility) presented significant improvements compared to the diabetic group (p<0.05). Conclusion: It seems that HIIT training can improve sperm count, survival rate and motility, through increasing serum testosterone, LH and FSH levels (reproductive hormones) in rats with diabetes mellitus type 2. The aim of this research is to estimate the effect and safety in application of betadine as a cheap and available chemical factor in treatment of pleurodesis. Materials and Methods: In this semi-empirical study, all patients suffering from recurrent pleural effusion enrolled the study. In order to perform pleurodesis, after chest tube insertion, solution containing iodopovidon, lidocaine, and normal saline was injected into the pleural cavity through chest tube. All the patients were examined for the pleural effusion and pleurodesis recurrence during a 6-month follow-up. Results: In this study, a total number of patients was 50 among whom 23 were male (46%) and 27 were female (54%). In the 6-month follow-up, 40 patients (80%) were completely recovered and the recurrence of the pleural effusion was noticed in 10 patients (20%). Comparison of the success rate of recovery in terms of the gender of the patients showed no significant relation between gender and response to the recovery process (p=0.219). Of the total of 32 patients (64%) with chest pain after pleurodesis, 23 patients had complete recovery, and the incidence of chest pain in patients with no improvement was significantly higher (p=0.018). Conclusion: Iodopovidone is a cheap, highly available material and of high efficacy in comparison to other sclerosant chemical substances, that provides an appropriate option in treatment of malignant pleural effusion.
