Abstract

  Background: Oxidative stress and severe neuro-excitation have significant effects on pathogenesis of Parkinson’s disease and agents with antioxidant property can potentially prevent these effects. Herein we examined potential protective effects of melatonin as an antioxidant agent and memantine as an uncompetitive receptor of NMDA, on a model of Parkinson’s disease induced by 1-methyl-4-phenyl-1, 2, 3, 6-tetrahydropyridine (MPTP).

  Materials and Methods: Male mice were divided into 8 groups with 7 mice in each group: saline, ethanol, melatonin, memantin, MPTP, melatonin+MPTP, memantin+ MPTP, melatonin+ memantin+ MPTP. All of agents were injected intraperitoneally once a day for 14 days before beam traversal test. Dopaminergic neurons of the Substantia Nigra Pars compacta (SNPC) were determined by immunohistochemical and were counted.

  Results: Melatonin improved notably movement dysfunction resulted of MPTP such as the number of errors, paces and the time of movement during behavioral test and also the counting of neurons of Substantia Nigra Pars Compacta. Memantin had a synergic effect on the most of improvements. However, the level of improvement and retrieval of signs was not as in saline and ethanol groups.

Conclusion: Melatonin especially together with memantine is able to prevent some of the MPTP-induced dysfunctions. However, the protective effects were not enogh, probably because of the amount of dose and the time of injection.

Abstract

Background: Toxocariasis is a parasitic disease caused by presence of Toxocara larva in human body. Soil as a reservoir maintained eggs of parasite and has important role in dissemination of disease because dogs and cats defecate eggs of parasite by feces. Despite there are large number of stray cats and dogs in urban areas such as Arak city, we need to collect information about soil pollution in public places. Therefore, the aim of this study was to evaluate soil contamination with Toxocara eggs in public parks in Arak city.

Materials and Methods: In this cross-sectional study, 60 soil samples were collected from 15 public parks in the Arak city during the summer 2015. The soil samples were provided by flotation method and examined by microscope.

Results: The results of this study showed that 4 (26.6%) of 15 parks were infected with Toxocara eggs. The highest rate of infection was observed in the walking and picnic site of parks. Also, in this study, two contaminated parks were located in the center and two parks were located in the southeastern of city.

Conclusion: Considering that soil contamination with Toxocara eggs was high in Arak city, it seems that the health authorities should design and implement programs in order to prevent human infection.

Abstract

Abstract
Background: Oxidative Stress and mitochondrial dysfunction leading to apoptotic death of neurons play key role in the pathogenesis of the Parkinson's disease. On the other hand، misregulation of microRNAs can cause several neurodegenerative diseases. Sirt1 and Bcl2 as two key genes, regulate pathogenic processes in neurodegenerative diseases such as mitochondrial dyfunction, oxidative stress and apoptosis.
 Materials and Methods: To investigate the role of microRNAs in model of Parkinson's disease, miRWalk 2.0 and TargetScan (v7) databases were served to predict microRNA-target interactions.
Results: Possible targeting effects of different microRNAs on Bcl2 and Sirt1 genes in Rat organism were analyzed. Merging data from databases has shown that rno-miR-449a, rno-miR-182, rno-miR-211, rno-miR-34b, rno-miR-34c, rno-miR-448, rno-miR-466b and rno-miR-96 with strong possibility can inhibit expression of Bcl2 gene. Also, rno-miR-181, rno-miR-211, rno-miR-27a, rno-miR-449a, rno-miR-34c, rno-miR-30, rno-miR-200a and rno-miR-448can inhibit Sirt1 gene with high possibility.
Conclusion: According to the findings, it can be predicted that regarding to high interaction scores of rno-miR-211, rno-miR-34c and rno-miR-448 and 449awith Bcl2 and Sirt1 genes in above-mentioned databases, these microRNAs probably can have critical role in disease process. Thus, these microRNAs can be introduced as appropriate candidates for investigations in in vitro model of Parkinson's disease.

Abstract

Background and Aim: Recent findings in cell therapy have presented new perspectives and opportunities for the treatment of neurodegenerative disease. The experimental research with intranasal (IN) administration of Stem Cells in Parkinson’s disease (PD) mouse model can work and in some cases induce major, long-lasting improvement. Adult Human endometrial derived stem cells (HEDSCs), a readily obtainable type of mesenchymal stem-like cell were used to generate dopaminergic cells and for cell therapy. In this study, we investigated the therapeutic effects of IN-delivered HEDSCs in mice model of Parkinson.
Materials and Methods: In this experimental research, 35 male mouse weighting 25-30 g were divided into 5 groups. On day 120 post cell administration, the rotational behavior was measured. Immunohistochemistry staining was used to detect HEDSCs in mice brain.
Findings: IN application of HEDSCs resulted in the appearance of cells in the substantia nigra (SN) and decrease in the rotational behavior of case group.
Conclusion: HEDSCs are a highly inducible source of allogenic stem cells that improve Parkinson’s disease.
